GHRP-6 (Growth Hormone Releasing Peptide-6) is a synthetic hexapeptide with the sequence His-D-Trp-Ala-Trp-D-Phe-Lys-NH2, a molecular weight of 873.0 g/mol, and CAS number 87616-84-0. It was among the first synthetic growth hormone secretagogues discovered and acts as a potent agonist of the ghrelin/growth hormone secretagogue receptor (GHSR1a).
Research demonstrates GHRP-6 stimulates growth hormone release from the anterior pituitary through direct activation of GHSR1a, a mechanism distinct from and synergistic with GHRH receptor activation. Studies show it suppresses somatostatin release while simultaneously stimulating GHRH release from the hypothalamus, producing robust GH pulses that exceed the response to GHRH alone.
Preclinical research documents GHRP-6’s orexigenic (appetite-stimulating) effects through central ghrelin receptor activation in the arcuate nucleus, distinguishing it from more selective secretagogues like ipamorelin. Studies also show GHRP-6 influences cortisol and prolactin release, increases gastric motility, and enhances gastric acid secretion through vagal afferent pathways.
