Research has established MOTS-c as a key regulator of metabolic homeostasis. Studies demonstrate it activates AMPK (AMP-activated protein kinase), the master cellular energy sensor, and modulates the folate-methionine cycle through inhibition of the de novo purine biosynthesis pathway. This leads to accumulation of AICAR, an endogenous AMPK activator, creating a metabolic signaling cascade that influences glucose uptake, fatty acid oxidation, and mitochondrial biogenesis.
In preclinical models, MOTS-c administration has been studied in models of age-dependent and high-fat diet-induced insulin resistance, physical performance capacity in aged mice, and cellular stress response pathways.
