MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA Type-c) is a mitochondria-derived peptide consisting of 16 amino acids with the sequence MRWQEMGYIFYPRKLR. It has a molecular weight of 2174.6 g/mol and CAS number 1627580-64-6. MOTS-c is encoded within the mitochondrial 12S rRNA gene and represents a class of signaling molecules known as mitochondrial-derived peptides (MDPs).

Research has established MOTS-c as a key regulator of metabolic homeostasis. Studies demonstrate it activates AMPK (AMP-activated protein kinase), the master cellular energy sensor, and modulates the folate-methionine cycle through inhibition of the de novo purine biosynthesis pathway. This leads to accumulation of AICAR, an endogenous AMPK activator, creating a metabolic signaling cascade that influences glucose uptake, fatty acid oxidation, and mitochondrial biogenesis.

In preclinical models, MOTS-c administration has been studied in models of age-dependent and high-fat diet-induced insulin resistance, physical performance capacity in aged mice, and cellular stress response pathways. Studies also reveal its translocation to the nucleus under metabolic stress, where it regulates gene expression through interaction with antioxidant response elements (ARE).
